Sterile Barrier System Testing for Medical Devices

1. Basic Concepts

Sterile Barrier System (SBS): As defined in ISO 11607, the minimum package that prevents ingress of microorganisms and allows aseptic presentation of the product at the point of use.

Related concepts:

  • Primary packaging: packaging in direct contact with the product

  • Sterile barrier system (SBS): the smallest packaging unit able to maintain sterility (e.g. blister tray + medical paper/Tyvek lid, paper-plastic pouch)

  • Protective packaging: outer packaging that prevents damage to the SBS during transport and storage

  • Packaging system: the combination of SBS + protective packaging

2. Key Regulations and Standards

Standard Content
ISO 11607-1 Requirements for materials, sterile barrier systems and packaging systems for terminally sterilized medical devices
ISO 11607-2 Validation requirements for forming, sealing and assembly processes
ASTM F1980 Standard guide for accelerated aging of sterile barrier systems
ASTM F1929 Detection of seal leaks by dye penetration
ASTM F88/F88M Seal strength of flexible barrier materials
ASTM F1140 Burst testing of flexible packages by internal pressurization
ASTM F2096 Detection of gross leaks by the bubble method
EN 868 series Packaging material requirements for terminally sterilized medical devices

3. Categories of Tests

1. Packaging material performance tests

Item Test method/standard Notes
Basis weight ISO 536 Mass per unit area of the material
Thickness ISO 534 Affects barrier performance
Tensile strength ASTM D882 / ISO 1924 Tensile properties of the material
Tear strength ASTM D1922 Tear resistance
Air permeability (Gurley) ASTM D726 Assesses suitability for EO sterilization
Microbial barrier ASTM F1608 Ability to block bacteria – the most critical property

2. Seal integrity tests

Item Standard Purpose
Dye penetration ASTM F1929 / YY/T 0681.5 Detects seal channel leaks (≥50 μm)
Bubble test ASTM F2096 / YY/T 0681.3 Detects gross leaks
Vacuum decay ASTM F2338 Non-destructive leak testing
Visual inspection ASTM F1886 Checks for visual seal defects

3. Seal strength tests

Item Standard Notes
Peel strength ASTM F88 / YY/T 0681.2 Measures the peel force of the seal
Burst test ASTM F1140 / F2054 Measures the pressure resistance of the whole package

4. Aging tests

  • Accelerated aging (ASTM F1980): uses the Arrhenius equation to simulate storage at room temperature by raising the temperature

    • Formula: AAT = RT × Q₁₀^((T_AA – T_RT)/10)

    • Common conditions: 55 °C to simulate shelf life at 25 °C, with Q₁₀ usually set to 2.0

    • Example: about 59 days at 55 °C ≈ 1 year at 25 °C

  • Real-time aging: run in parallel with accelerated aging; real-time data ultimately prevails

5. Distribution simulation tests

Item Standard
Vibration ASTM D4169 / ISTA 2A / ISTA 3A
Drop ASTM D5276
Stacking/compression ASTM D4577
Temperature and humidity cycling ASTM D4332
Low pressure (air transport) ASTM D6653

4. Typical Test Flow

Sample preparation → sterilization → accelerated aging + real-time aging (started together)

Key time points: verification is usually performed at three points: month 0, the end of the accelerated-aging equivalent shelf life, and the end of the actual real-time aging period.

5. Considerations for Different Package Types

Package type Common combinations Test focus
Flexible–rigid Blister tray + Tyvek lid Lid peel force, seal uniformity, microbial barrier
Flexible–flexible Paper-plastic pouches, four-side-seal pouches Peel strength, heat-seal channels
Rigid–rigid Glass vials + rubber stoppers, prefilled syringes Container closure integrity (CCI)

6. FAQs and Points to Note

  1. Sample size: usually at least 30 samples per test and per time point; for critical tests, determined by AQL or statistical requirements

  2. Effect of sterilization: EO, irradiation and moist heat affect packaging materials differently; sterilization should be completed before testing

  3. Shelf-life claims: must be supported by real-time aging data; accelerated aging only provides interim support for early market launch

  4. Change management: revalidation is required after changes to packaging materials, suppliers, heat-seal parameters or sterilization method

  5. Process validation (ISO 11607-2): includes IQ/OQ/PQ; heat-seal parameters must be process-validated
